An important aspect of predictive processing is that each hypothesis generated by a level in the hierarchy is associated with a notion of confidence in the hypothesis, which in turn is based on prior expectations. Could psychedelics be altering our perception of reality by messing with this process? Friston and Robin Carhart-Harris think so. If psychedelics mess with prior beliefs, that might also explain why they cause one to hallucinate a reality that’s untethered from real-world expectations.

Semaglutide
The mean change in body weight from baseline to week 68 was −14.9% in the semaglutide group as compared with −2.4% with placebo, for an estimated treatment difference of −12.4% (95% confidence interval [CI], −13.4 to −11.5; P<0.001). More participants in the semaglutide group than in the placebo group achieved weight reductions of 5% or more (1047 participants [86.4%] vs. 182 [31.5%]), 10% or more (838 [69.1%] vs. 69 [12.0%]), and 15% or more (612 [50.5%] vs. 28 [4.9%]) at week 68 (P<0.001 for all 3 comparisons of odds). The change in body weight from baseline to week 68 was −15.3 kg in the semaglutide group as compared with −2.6 kg in the placebo group (estimated treatment difference, −12.7 kg; 95% CI, −13.7 to −11.7). Participants who received semaglutide had a greater improvement with respect to cardiometabolic risk factors and a greater increase in participant-reported physical functioning from baseline than those who received placebo.
this could be the first weight loss drug that actually works. if true, huge public health implications.
Dead drop drugs
Most Russian drug dealers don’t hand off drugs anymore—they stash them in geotagged hiding spots ready for pickup by online buyers.
Drug Experiences
US Army $1b LSD Stash
we should probably talk about the time that Ketchum found $1b worth of LSD, enough to “intoxicate several 100m people”, just sitting in his office—and the fact that it disappeared without explanation.
500kg of Cocaine
Quinci’s arrival on São Miguel had changed the island in surprising ways. A number of locals had become rich thanks to the Italian’s cocaine, then started legitimate businesses, such as coffee shops, many of which still exist today. But the drugs also had more damaging long-term effects. Quinci’s cocaine was so potent that they started taking other drugs to lessen the symptoms of withdrawal. They became addicted to heroin, which was shipped in from the continent, often via the postal service. The arrival of Quinci’s cocaine increased consumption of other illicit substances, and young people and adults from poorer parts of the island were the ones most affected. “It completely ruined my life. I’m still living with consequences to this day.”
Bernie und Ert
Bernie und Ert was a recurring sketch on the show created by Attik Kargar, who performed the puppets and supplied the voice of Bernie. Bernie and Ert are an obscene parody of Bert and Ernie on Sesame Street, and especially as re-dubbed on Sesamstrasse. The pair are a same-sex couple, with no nose and one eye each. Each sketch focused on such topics as crime, drug abuse (Bernie is a cocaine addict), and S&M practices.
Crack pipe vending crackdown
3 vending machines dispensing crack pipes for $2 each were discovered roadside in Long Island. The machines were marked “PENS” and did indeed contain pens, well, ballpoint pens that had been turned into crack pipes. Town of Brookhaven Supervisor Ed Romaine tested the machine by inserting the required 8 quarters and later remarked, “We’re going to crack down on this.”
Viagra
But while Viagra is poised to go wider than ever before, the inside story of its launch is not widely known. How did a group of oddball underdogs in America’s most conservative pharmaceutical conglomerate, Pfizer, bring it into existence? At the time, the idea of selling Viagra was considered crazy at best and immoral at worst.
In fact, it’s a miracle that it ever came to be at all. In addition to the people within Pfizer who were in an uproar over the “dick pill,” 4 major groups began rallying against it before its launch: the Catholic church (which thought it was immoral), medical experts (who insisted patients would be too embarrassed to ask for the pill), business execs (who thought it would make Pfizer a laughingstock), and legislators (who lobbied against the pill for the same reason as the church).
Melatonin
Van Geiklswijk et al describe supplemental melatonin as “a chronobiotic drug with hypnotic properties”. Using it as a pure hypnotic – a sleeping pill – is like using an AK-47 as a club to bash your enemies’ heads in. It might work, but you’re failing to appreciate the full power and subtlety available to you.
the correct dose is 0.3 mg:
Most drugstore melatonin supplements are 10x or more the recommended dose. the FDA chose to label it a dietary supplement, which does not require FDA regulation. Clearly, this was wrong because melatonin is a hormone, not a dietary supplement. Quickly, supplement manufacturers saw the huge potential in selling melatonin to promote good sleep. After all, millions of Americans struggled to get to sleep and stay asleep, and were desperate for safe alternatives to anti-anxiety medicines and sleeping pills that rarely worked well and came with plenty of side effects. Also, manufacturers must have realized that they could avoid paying royalties to MIT for melatonin doses over the 1 mg measure. So, they produced doses of 3 mg, 5 mg, 10 mg and more! Their thinking–like so much else in our American society–was likely, “bigger is better!” But, they couldn’t be more wrong.
